The last league meeting was a 2-2 draw at Somerset Park in March.
Ayr ran out 2-0 winners against City in the League Cup first round on August 1.
Ian McCall's men suffered a 3-0 defeat at Albion Rovers last weekend.
Darren Dods' side were beaten 6-1 by Dunfermline in their first league game.
Dunfermline Athletic (1st) v Cowdenbeath (7th)
Dunfermline beat the Blue Brazil 5-1 in the first round of the League Cup.
The Pars have also won 4-1 at Arbroath and 6-1 at Brechin this season.
Cowdenbeath were beaten 2-1 at home by Stranraer in their opening league game.
Cowdenbeath won 3-0 in the play-offs at East End Park in May 2014.
Forfar Athletic (4th) v Albion Rovers (2nd)
Forfar kicked off their League 1 campaign with a 1-0 win at Airdrieonians.
The teams last met in the league in 2012/13 - Forfar twice won 4-2 at Station Park.
Albion Rovers beat Ayr United 3-0 at Cliftonhill in their first league game.
Darren Young's side finished 10 points clear of second-placed Queen's Park last season.
Stenhousemuir (6th) v Airdrieonians (8th)
Stenhousemuir drew 2-2 at Peterhead last week after being 2-0 up early on.
Airdrie lost 1-0 to Forfar at the Excelsior Stadium, Chris Templeman scoring in two minutes.
The Diamonds won 2-1 at home when the sides last met at the end of March.
Gary Bollan's men have won the last three meetings between the teams.
Stranraer (3rd) v Peterhead (5th)
It's a distance of 263 miles from Balmoor to Stair Park - a five-hour journey.
Stranraer won all five league and cup meetings between the teams last season.
Peterhead's last victory at Stair Park was 3-0 in April 2012.
Craig Malcom scored a last-minute winner in Stranraer’s 2-1 win at Cowdenbeath last week.
Peterhead fought back from two goals down to draw 2-2 with Stenhousemuir last week.
